Brazil's Silva beginning chemotherapy for tumor

(AP) ? The tumor found in the throat of Brazil's popular former President Luiz Inacio Lula da Silva was detected in an intermediate stage and shows the "classic aggressiveness" of larynx cancer, doctors said Monday.

Silva entered Sao Paulo's Sirio-Libanes Hospital to undergo his first chemotherapy treatment Monday. The team of physicians treating him said he was upbeat and they were confident he would make a full recovery. Silva is expected to also undergo radiation therapy early next year, and doctors were hopeful surgery to remove the tumor would not be necessary.

"It's the most common type of tumor in this part of the body, and it has a classic aggressiveness," said Dr. Paulo Hoff, one of the oncologists overseeing Silva's treatment. "The tumor was detected in an intermediate stage, relatively early but not so early it could be resolved with a small surgery."

The cancer had not spread to other parts of Silva's body, Hoff said. He confirmed that because of the chemotherapy, Silva, 66, would lose his hair and thick, gray beard, a trademark look he maintained through his decades as a union leader who aggressively took on Brazil's military dictatorship and then through a long political career.

President Dilma Rousseff, who was elected last year because of Silva's tireless campaigning, was expected to visit him at the Sao Paulo hospital later Monday.

In 2009, Rousseff had a malignant tumor removed from her left armpit at the same hospital. She underwent chemotherapy treatment and was given a clean bill of health in August 2010.

Doctors said Silva would spend the night as a precautionary step, so they could monitor his response to the first chemotherapy treatment.

Dr. Gady Har-El, chairman of head and neck surgery at Lenox Hill Hospital in New York, said that the use of chemotherapy for Silva is an indication the cancer was not caught in its early stages.

"In general, early cancer of the larynx is treated with just one modality, either radiation or surgery," he said. "The fact they added chemotherapy says it's not an early cancer."

Har-El said that because of the chemotherapy, Silva can expect discomfort in the throat and difficulty in swallowing, the latter of which could lead to the his needing a feeding tube inserted into his stomach if the pain inhibits his ability to eat.

Silva's doctors said that because of the location of the tumor, surgery would carry the chance that Silva could lose his voice, a development that could have an impact on Brazilian politics, as Silva's rousing campaign rallies and high-energy speeches are a potent political weapon.

Silva maintains enormous influence within the ruling Workers Party that he founded, and Alexandre Barros, with the Early Warning political risk group in Brasilia, said that Rousseff's administration has yet to distinguish itself as being independent from Silva's legacy, so his continued support of the president is vital

Brazil will hold nationwide municipal elections next year, and candidates from the Workers Party may be hurt if Silva cannot attend rallies and participate in campaigns.

The next presidential election will be held in 2014. Despite speculation he might run himself, Silva has said Rousseff will be the ruling party's candidate.

"There is the possibility that if he survives, he could help win even more votes in 2014," Barros said. "If he were to lose his voice, it's hard to say what the result would be, though I've never heard of there being a voiceless leader in the world."

Silva's doctors said it will be about 40 days before they will know how Silva's tumor is responding to the chemotherapy.

Dr. Roberto Kalil, Silva's physician since he first won the presidency, said the former leader was upbeat when he entered the hospital and prepared for the challenge awaiting him.

"He's very calm. Obviously at first there was anxiety, a shock," Kalil said. "He arrived in excellent spirits, in his usual way, extremely confident."

Gaza violence simmers after truce announced (Reuters)

GAZA (Reuters) ? Violence between Israel and Gaza militants flared anew Sunday after Egypt said it had brokered a truce to end attacks that have killed 10 Palestinian gunmen and an Israeli civilian in the past five days.

A ceasefire had appeared to be taking hold, with the frontier quiet for some eight hours, until Israeli aircraft attacked Palestinian militants who the military said were about to fire rockets from the southern Gaza Strip.

Medical officials in the enclave said one man was killed and another wounded, identifying them as members of the Democratic Front for the Liberation of Palestine, a group that rarely launches attacks against Israel.

Islamic Jihad and two smaller factions have claimed responsibility for rocket strikes against Israel since Wednesday. Islamic Jihad has close ties with Iran and has chafed at the rule of rival Hamas Islamists in the Gaza Strip.

An Islamic Jihad source, speaking several hours after the ceasefire was announced by Egyptian officials, said the group welcomed Cairo's efforts.

"If the (Israeli) aggression is stopped, we will abide by the calm," said Abu Ahmed, leader of Islamic Jihad's armed wing said.

Israel and Palestinian militants have used such language in the past in ceasing fire while stopping short of acknowledging any formal truce agreement.

"If someone rises up to kill you, kill him first," Israeli Prime Minister Benjamin Netanyahu said in public remarks several hours before the latest air strike.

But he said Israel was "not eager for escalation."

DEADLY ATTACKS

The current round of cross-border violence began when residents of Israeli suburbs in the southern approaches to Tel Aviv were jolted awake Wednesday night by sirens usually sounded in towns and villages closer to the Gaza Strip.

The alert was triggered by what Israeli security sources said was an upgraded long-range Grad rocket that struck harmlessly near the Israeli port of Ashdod, leading to an Israeli air strike Saturday that killed five top Islamic Jihad militants in a Gaza training camp.

Five other militants have died in subsequent air attacks.

One of some 30 rockets and mortar bombs fired at Israel killed an Israeli man in the city of Ashkelon. Two other people were wounded.

"My interpretation is that these guys (Islamic Jihad) are keen to show off their clout, which has been built up recently," Yosef Kupperwasser, director-general of Israel's Strategic Affairs Ministry, told Reuters.

Israel kept schools shut in its southern region Sunday as a precaution against further rocket fire, while hundreds of thousands of civilians within 40 km (25 miles) of Gaza were urged to stay indoors.

Islamic Jihad released images of what it said was the firing by its men of a truck-mounted multiple rocket-launcher, a platform not previously seen in Gaza.

Israel says Gaza arsenals have been boosted by gun-running from Libya since the fall of its ruler, Muammar Gaddafi.

The violence erupted after weeks of a relative lull surrounding a prisoner swap on October 18 in which Israel released 477 Palestinian prisoners in exchange for Gilad Shalit, an Israeli soldier held by Hamas since 2006.

(AP) ? If Will and Kate's first child is a girl, it's now clear that she'll probably become queen one day ? and not even getting a little brother can mess that up.

The Commonwealth countries agreed Friday to change centuries-old rules of succession that put sons on the throne ahead of any older sisters. So that hypothetical daughter of Prince William and Kate Middleton ? now known as Catherine, Duchess of Cambridge ? would have a prime place in history: the first princess to beat out any younger brothers and accede to the throne.

Had these rules been in place in the 1500s, Henry VIII would have just been a rather large historical footnote.

The move is a baby step: Before taking effect, the changes still must be approved by the legislatures of the 16 nations where Queen Elizabeth II is head of state. Still, the agreement, which was reached at a meeting of Commonwealth nations in Perth, Australia, represents a triumph over practices now considered outdated and sexist in much of the world.

Nations including Sweden, Belgium, the Netherlands and Norway have already taken similar steps.

Will and Kate's lavish April wedding renewed a decades-long debate over succession.

Middleton told a well-wisher in Canada this summer that she hopes to start a family. William has said the same.

Once their honeymoon was over, baby talk started, adding urgency to the dialogue, although officials insist that talk of a pregnancy is premature.

Historians think it's about time.

"You shouldn't muck around too much with the constitution, but it's a good idea to change this at this time," said royal expert Hugo Vickers. "It's much better to have it sorted out before any babies come along."

The new rules would only apply to future heirs and would have no impact on the current line of succession.

William is second in line to the throne after his father, Prince Charles, who is the queen's firstborn child. Charles' sister, Anne, is lower in the line of succession than her younger brothers Andrew and Edward by virtue of their male gender.

Charles had only sons, William and Prince Harry, so the issue of gender was never raised.

In 2009, the government of then-Prime Minister Gordon Brown considered a bill that would end the custom of putting males ahead of females in the succession line. It also would lift a ban on British monarchs marrying Roman Catholics. The government did not have time to pursue it before Brown left office.

The rule has kept women from succeeding to the throne in the past. Queen Victoria's first child was a daughter ? also called Victoria ? but it was her younger brother who became King Edward VII.

If Queen Victoria had been able to pass her crown to her firstborn, Britain's Princess Victoria would have had a brief reign before her death in 1901.

That would have made her son ? Wilhelm II, who at that time was the German Kaiser ? king. With Wilhelm II ruling both Germany and Britain, there may not have been two world wars.

Earlier history might also have been drastically different if women had had equal rights to the throne.

Neither Henry VIII nor Charles I would have been king because both had older sisters who, under the new rules, would have been monarch.

As king, Henry VIII set in motion the creation of the Church of England. His six marriages left an insecure succession ? one sickly son and two princesses, according to the monarchy's official website. Charles I's reign in the 17th century led to a bloody civil war.

Prince William and his wife have been credited with freshening up a staid monarchy, and new succession rules seem to fit right in.

"In this day and age, why should a royal son be more important than a royal daughter?" said Joe Little, managing editor of Majesty magazine.

The same goes for the decision Friday in Perth to lift a ban on monarchs marrying Roman Catholics. Critics had called the rule blatantly discriminatory since royals are free to wed Jews, Muslims, Hindus or members of any other religion."Britain is no longer the religious country that it once was," Little said. "While not denigrating the importance of religion, it plays much less of a role now then it did 60 years ago."

Still, some Britons are wary of a Catholic monarch.

"The pope is responsible for some horrors," said Anna Marsh, 73, who was cycling in London.

Her biking buddy Jill Gregory, 71, was fine with the idea ? and also fully in favor of giving firstborn girls an equal right to the throne.

"In terms of ability, I don't think women are any different than men," Gregory said, pointing to the queen and her late mother.

Elizabeth II succeeded her father, King George VI, because he had no sons. If she had had a younger brother, he would have jumped above her in the line of succession.

Prime Minister David Cameron had pushed for the changes, calling it a matter of equality.

New Zealand will now chair a working group of Commonwealth countries to discuss how to accomplish the reforms. It's not a simple process. Getting all 16 countries to begin the legislative changes is what has held them up for decades.

However long it takes, Patricia Wager of London said it would clear up something that should not be an issue in the modern world.

"It's a good idea, and a long time coming," she said.

(AP) ? Anti-Wall Street demonstrators held vigils for an Iraq War veteran seriously injured during a protest clash with police in California as some Occupy encampments came under growing pressure from authorities to abandon sites in parks and plazas.

A crowd of at least 1,000 people, many holding candles, gathered Thursday night in Oakland in honor of 24-year-old Scott Olsen, who is hospitalized with a fractured skull.

Many in the crowd shooed away Oakland Mayor Jean Quan who retreated back into City Hall after trying to address them during a tense late-night appearance. She apologized to Olsen during a hospital visit earlier Thursday.

"I am deeply saddened about the outcome on Tuesday. It was not what anyone hoped for, ultimately it was my responsibility, and I apologize for what happened," Quan said in a written statement to protesters late Thursday. "I cannot change the past, but I want to work with you to ensure that this remains peaceful moving forward."

In Nashville, police cracked down overnight on an Occupy protest camp near the Capitol under a new policy setting a curfew for the complex. Officers moved in a little after 3 a.m. and arrested about 30, who were later released after a judge wouldn't sign the warrants. About 20 protesters who stayed on a nearby sidewalk were not arrested and were still there later in the morning as state troopers stood guard at the steps to the Capitol.

Protesters also held a vigil for Olsen in Las Vegas, which drew a handful of police officers. Afterward, protesters invited them back for a potluck dinner.

"We renewed our vow of nonviolence," organizer Sebring Frehner said.

The Marine veteran, who won medals in Iraq, has become a rallying cry for the Occupy Wall Street demonstrators across the nation, with Twitter users and protest websites declaring, "We are all Scott Olsen."

Joshua Shepherd, 27, a Navy veteran who was standing nearby when Olsen got struck, called it a cruel irony that Olsen is fighting an injury in the country that he fought to protect.

Despite the financial underpinnings of the protests, Olsen himself wasn't taking part out of economic need.

His friends say he makes a good living as a network engineer and has a nice apartment overlooking San Francisco Bay. Still, he felt so strongly about economic inequality in the United States that he fought for overseas that he slept at a protest camp after work.

"He felt you shouldn't wait until something is affecting you to get out and do something about it," said friend and roommate Keith Shannon, who served with Olsen in Iraq.

It was that feeling that drew him to Oakland on Tuesday night, when the clashes broke out and Olsen's skull was fractured. Fellow veterans said Olsen was struck in the head by a projectile fired by police, although the exact object and who might have been responsible for the injury have not been definitively established. Officials are investigating exactly where the projectile came from.

Even as the vigil was held in Oakland, protest organizers prepared to defy Oakland's prohibition on overnight camping on the now patchy, manure-smelling lawn outside City Hall.

Shake Anderson, an organizer with Occupy Oakland, said half a dozen tents were erected on the plaza by midday Thursday where police armed with tear gas and bean bag rounds disbanded a 15-day-old encampment Tuesday. More than two-dozen tents had been erected as food and supplies arrived late Thursday.

"We believe in what we're doing," Anderson said. "No one is afraid. If anything, we're going to show there's strength in numbers."

Few police were seen in the area during late Thursday, as Quan in her written statement said that she and interim police chief Howard Jordan hope to meet with protesters and urged them again not to camp at the plaza.

Elsewhere across the United States, protesters brushed off pressure from authorities and maintained the camps that have sprung up in opposition to growing economic inequality.

Protesters at San Francisco's Justin Herman Plaza braced for a police raid early Thursday that never came. Still, police have warned the protesters that they could be arrested on a variety of sanitation or illegal camping violations.

Officials told protesters in Providence, R.I., that they were violating multiple city laws by camping overnight at a park.

Anti-Wall Street protesters camped out in downtown Los Angeles said they're planning to continue their demonstration indefinitely, although both they and the mayor's office were eyeing alternate sites.

Meanwhile, Olsen has been improving. Doctors transferred him from the emergency room to an intensive care unit and upgrading his condition to fair on Thursday.

Dr. Alden Harken, chief surgeon at Highland Hospital, said Olsen was still unable to speak but had improved dramatically since he was hospitalized unconscious with a fractured skull and bruised brain that caused seizures.

Harken said Olsen was interacting with his parents, who flew in from Wisconsin, doing math equations and otherwise showing signs of "high-level cognitive functioning." The doctor said he may require surgery, but that's unlikely.

"He's got a relatively small area of injury and he's got his youth going for him. So both of those are very favorable," Harken said.

Olsen smiled during Quan's visit and expressed surprise at all the attention his injury has generated, hospital spokesman Vintage Foster said.

His uncle in Wisconsin told The Associated Press that Olsen's mother was trying to understand what had happened.

"This is obviously a heartbreaker to her," George Nygaard said. "I don't think she understands why he was doing this."

The group Iraq Veterans Against the War blamed police for Olsen's injury. Jordan said the next day that Oakland police will investigate whether officers used excessive force.

Police have said they responded with tear gas and bean bag rounds only when protesters began throwing bottles and other items at them.

On Tuesday, Olsen had planned to be at the San Francisco protest, but he changed course after his veterans' group decided to support protesters in Oakland after police cleared a two-week long encampment outside City Hall.

"I think it was a last-minute thing," Shannon said.

A video posted on YouTube showed Olsen being carried by other protesters through the tear gas, his face bloodied. People shout at him: "What's your name? What's your name?" Olsen just stares back.

People at OPSWAT, the San Francisco security software company where Olsen works, were devastated after learning of his injuries. They described him as a humble, quiet man.

Olsen had been helping to develop security applications for U.S. defense agencies, building on expertise gained while on active duty in Iraq, said Jeff Garon, the company's director of marketing.

Olsen was awarded seven medals while serving in the U.S. Marine Corps, which he left as a lance corporal in November 2009 after serving for four years. One of them was the Navy-Marine Corps Achievement Medal.

Olsen moved to the Bay Area in July, and quickly found friends in the veterans against the war group.

His tours of duty in Iraq made him more serious, Shannon said.

"He wasn't active in politics before he went in the military, but he became active once he was out ... the experience in the military definitely shaped him," Shannon said.

Pink Floyd member's son loses jail term appeal (AP)

LONDON ? Appeal court judges on Friday upheld the 16-month jail sentence given to the son of Pink Floyd guitarist David Gilmour for a violent rampage during student protests last year.

Charlie Gilmour was one of thousands who demonstrated in December against rising university tuition fees and was among a group that broke away from the main demonstration and attacked a convoy carrying Prince Charles and his wife, Camilla. At Gilmour's trial earlier this year, a judge said Gilmour had jumped on the hood of a Jaguar in the royal motorcade and thrown a garbage can at the car.

Gilmour also kicked a store window, stole the leg of a mannequin and was photographed hanging from a Union flag on the Cenotaph, a memorial to British war dead.

The 21-year-old Cambridge University student, who has been in jail since July, pleaded guilty to violent disorder but challenged the length of his sentence.

But three appeals judges said the sentence was neither "manifestly excessive (nor) wrong in principle."

"We do not believe that violence in this context and of the kind displayed by this defendant can normally be met by other than significant sentences of immediate custody even for those of otherwise good character," said one of the three, Anthony Hughes.

Big dinos hit the road when they needed food

What did giant plant-munching dinosaurs do when they couldn't find enough to eat in the parched American West? They hit the road.

An analysis of fossilized teeth adds further evidence that the long-necked dinosaurs called sauropods ? the largest land creatures ? went on road trips to fill their gargantuan appetites.

Scientists have long theorized that sauropods foraged for precious resources during droughts because of their preserved tracks and long limbs that were "ideal moving machines" and allowed them to cover long distances, said paleobiologist Matthew Bonnan of Western Illinois University.

The latest study is the best evidence yet that at least one kind of sauropod "took to the hills in search of food when times got tough in the lowlands," said paleontologist Kristi Curry Rogers at Macalester College in Minnesota.

The new work, published online Wednesday by the journal Nature, was led by geologist Henry Fricke of Colorado College.

The researchers analyzed 32 sauropod teeth collected in Wyoming and Utah. The teeth came from massive plant-eaters that roamed a semi-arid basin in the American West during the late Jurassic period about 150 million years ago.

Scientists can get a glimpse into the source of the dinosaurs' drinking water by comparing the oxygen preserved in the tooth enamel to that found in ancient sediment.

A chemical analysis showed differences in the teeth and the basin where the dinosaurs were buried, meaning they must have wandered hundreds of miles from the flood plains to the highlands for food and water.

Fricke said the movement appeared to be tied to changing seasons. Sauropods left the basin in the summer for higher elevations ? a trek that took about five months ? and returned in the winter.

In lush times, sauropods would have feasted on a diversity of plants including ferns, horsetails, conifers and moss, said John Foster, a curator at the Museum of Western Colorado, who had no part in the research.

As the Poetaster, you get to be the boss man, controlling others just by speaking, owning a house, so on... In such, I do need a rather trust-able player to play him. But he should be a fun role.

The singer is the Poetaster's right hand man. He came around second to the band, in some sort of freak accident where the Poetaster learned about his ability to control other people.

Plants feel the force

"Picture yourself hiking through the woods or walking across a lawn," says Elizabeth Haswell, PhD, assistant professor of biology in Arts & Sciences at Washington University in St. Louis. "Now ask yourself: Do the bushes know that someone is brushing past them? Does the grass know that it is being crushed underfoot? Of course, plants don't think thoughts, but they do respond to being touched in a number of ways."

"It's clear," Haswell says, "that plants can respond to physical stimuli, such as gravity or touch. Roots grow down, a 'sensitive plant' folds its leaves, and a vine twines around a trellis. But we're just beginning to find out how they do it," she says.

In the 1980s, work with bacterial cells showed that they have mechanosensitive channels, tiny pores in the cells membrane that open when the cell bloats with water and the membrane is stretched, letting charged atoms and other molecules to rush out of the cell. Water follows the ions, the cell contracts, the membrane relaxes, and the pores close.

Genes encoding seven such channels have been found in the bacterium Escherichia coli and 10 in Arabidopsis thaliana, a small flowering plant related to mustard and cabbage. Both E. coli and Arabidopsis serve as model organisms in Haswell's lab.

She suspects that there are many more channels yet to be discovered and that they will prove to have a wide variety of functions.

Recently, Haswell and colleagues at the California Institute of Technology, who are co-principal investigators on an National Institutes of Health (NIH) grant to analyze mechanosensitive channels, wrote a review article about the work so far in order to "get their thoughts together" as they prepared to write the grant renewal. The review appeared in the Oct. 11 issue of Structure.

Swelling bacteria might seem unrelated to folding leaflets, but Haswell is willing to bet they're all related and that mechanosensitive ion channels are at the bottom of them all. After all, plant movements ? both fast and slow ? are ultimately all hydraulically powered; where ions go the water will follow.

Giant E. coli cells

The big problem with studying ion channels has always been their small size, which poses formidable technical challenges.

Early work in the field, done to understand the ion channels whose coordinated opening and closing creates a nerve impulse, was done in exceptionally large cells: the giant nerve cells of the European squid, which had projections big enough to be seen with the unaided eye.

Experiments with these channels eventually led to the development of a sensitive electrical recording technique known as the patch clamp that allowed researchers to examine the properties of a single ion channel. Patch clamp recording uses as an electrode a glass micropipette that has an open tip. The tip is small enough that it encloses a "patch" of cell membrane that often contains just one or a few ion channels.

Patch clamp work showed that there were many different types of ion channels and that they were involved not just in the transmission of nerve impulses but also with many other biological processes that involve rapid changes in cells.

Mechanosensitive channels were discovered when scientists started looking for ion channels in bacteria, which wasn't until the 1980s because ion channels were associated with nerves and bacteria weren't thought to have a nervous system.

In E. coli, the ion channels are embedded in the plasma membrane, which is inside a cell wall, but even if the wall could be stripped away, the cells are far too small to be individually patched. So the work is done with specially prepared giant bacterial cells called spherophlasts.

These are made by culturing E. coli in a broth containing an antibiotic that prevents daughter cells from separating completely when a cell divides. As the cells multiply, "snakes" of many cells that share a single plasma membrane form in the culture. "If you then digest away the cell wall, they swell up to form a large sphere," Haswell says.

Not that spheroplasts are that big. "We're doing most of our studies in Xenopus oocytes (frog eggs), whose diameters are 150 times bigger than those of spheroplasts," she says.

Three mechanosensitive channel activites

To find ion channels in bacteria, scientists did electrophysiological surveys of spheroplasts. They stuck a pipette onto the spheroplast and applied suction to the membrane as they looked for tiny currents flowing across the membrane.

"What they found was really amazing," Haswell says. "There were three different activities that are gated (triggered to open) only by deformation of the membrane." (They were called "activities" because nobody knew their molecular or genetic basis yet.)

The three activities were named mechanosensitive channels of large (MscL), small (MscS) and mini (MscM) conductance. They were distinguished from one another by how much tension you had to introduce in order to get them to open and by their conductance.

One of the labs working with spheroplasts was led by Ching Kung, PhD, at the University of Wisconsin-Madison. The MscL protein was identified and its gene was cloned in 1994 by Sergei Sukharev, PhD, then a member of Kung's lab. His tour-de-force experiment, Haswell says, involved reconstituting fractions of the bacterial plasma membrane into synthetic membranes (liposomes) to see whether they would confer large-channel conductance.

In 1999, the gene encoding MscS was identified in the lab of Ian Booth, PhD, at the University of Aberdeen. Comparatively, little work has been done on the mini channel, which is finicky and often doesn't show up, Haswell says, though a protein contributing to MscM activity was recently identified by Booth's group.

Once both genes were known, researchers did knockout experiments to see what happened to bacteria that didn't have the genes needed to make the channels. What they found, says Haswell, was that if both the MscL and MscS genes were missing, the cells could not survive "osmotic downshock," the bacterial equivalent of water torture.

"The standard assay," Haswell says, "is to grow the bacteria for a couple of generations in a very salty broth, so that they have a chance to balance their internal osmolyte concentration with the external one." (Osmolytes are molecules that affect osmosis, or the movement of water into and out of the cell.) "They do this," she says, "by taking up osmolytes from the environment and by making their own."

"Then," she says, "you take these bacteria that are chockfull of osmolytes and throw them into fresh water. If they don't have the MscS and MscL proteins that allow them to dump ions to avoid the uncontrolled influx of water, they don't survive." It's a bit like dumping saltwater fish into a freshwater aquarium.

Why are there three mechanosenstivie channel activities? The currently accepted model, Haswell says is that the channels with the smaller conductances are the first line of defense. They open early in response to osmotic shock so that the channel of large conductance, through which molecules the cell needs can escape, doesn't open unless it is absolutely necessary. The graduated response thus gives the cell its best chance for survival.

Crystallizing the proteins

The next step in this scientific odyssey, figuring out the proteins' structures, also was very difficult. Protein structures are traditionally discovered by purifying a protein, crystallizing it out of a water solution, and then bombarding the crystal with X-rays. The positions of the atoms in the protein can be deduced from the X-ray diffraction pattern.

In a sense crystallizing a protein isn't all that different from growing rock candy from a sugar solution, but, as always, the devil is in the details. Protein crystals are much harder to grow than sugar crystals and, once grown, they are extremely fragile. They even can even be damaged by the X-ray probes used to examine them.

And to make things worse MscL and MscS span the plasma membrane, which means that their ends, which are exposed to the periplasm outside the cell and the cytoplasm inside the cell, are water-loving and their middle sections, which are stuck in the greasy membrane, are repelled by water. Because of this double nature it is impossible to precipitate membrane proteins from water solutions.

Instead the technique is to surround the protein with what have been characterized as "highly contrived detergents," that protect them ? but just barely ? from the water. Finding the magical balance can take as long as a scientific career.

The first mechanosensitive channel to be crystallized was MscL?not the protein in E. coli but the analogous molecule (a homolog) from the bacterium that causes tuberculosis. This work was done in the lab of one of Haswell's co-authors, Douglas C. Rees a Howard Hughes investigator at the California Institute of Technology.

MscS from E. coli was crystallized in the Rees laboratory several years later, in 2002, and an MscS protein with a mutation that left it stuck in the presumed open state was crystallized in the Booth laboratory in 2008. "So now we have two crystal structures for MscS and two (from different bacterial strains) for MscL," Haswell says.

Of plants and mutants

Up to this point, mechanosensitive channels might not seem all that interesting because the lives of bacteria are not of supreme interest to us unless they are making us ill.

However, says, Haswell, in the early 2000s, scientists began to compare the genes for the bacterial channels to the genomes of other organisms and they discovered that there are homologous sequences not just in other bacteria but also in some multicellular organisms, including plants.

"This is where I got involved," she says. "I was interested in gravity and touch response in plants. I saw these papers and thought these homologs were great candidates for proteins that might mediate those responses."

"There are 10 MscS-homologs in Arabidopsis and no MscL homologs," she says. "What's more, different homologs are found not just in the cell membrane but also in chloroplast and mitochondrial membranes. "

The chloroplast is the light-capturing organelle in a plant cell and the mitochondria is its power station; both are thought to be once-independent organisms that were engulfed and enslaved by cells which found them useful. Their membranes are vestiges of their free-living past.

The number of homologs and their locations in plant cells suggests these channels do much more than prevent the cells from taking on board too much water.

So what exactly were they doing? To find out Haswell got online and ordered Arabidopsis seeds from the Salk collection in La Jolla, Calif., each of which had a mutation in one of the 10 channel genes.

From these mutants she's learned that two of the ten channels control chloroplast size and proper division as well as leaf shape. Plants with mutations in these two MscS channel homologs have giant chloroplasts that haven't divided properly. The monster chloroplasts garnered her lab the cover of the August issue of The Plant Cell.

"We showed that bacteria lacking MscS and MscL don't divide properly either,"Haswell says, "so the link between these channels and division is evolutionarily conserved."

The big idea

But Haswell and her co-authors think they are only scratching the surface. "We are basing our understanding of this class of channels on MscS itself, which is a very reduced form of the channel," she says. "It's relatively tiny."

"But we know that some of the members of this family have long extensions that stick out from the membrane either outside or inside the cell. We suspect this means that the channels not only discharge ions, but that they also signal to the whole cell in other ways. They may be integrated into common signaling pathways, such as the cellular osmotic stress response pathway.

We think we may be missing a lot of complexity by focusing too exclusively on the first members of this family of proteins to be found and characterized," she says. "We think there's a common channel core that makes these proteins respond to membrane tension but that all kinds of functionally relevant regulation may be layered on top of that."

"For example," she says, "there's a channel in E. coli that's closely related to MscS that has a huge extension outside the cell that makes it sensitive to potassium. So it's a mechanosensitive channel but it only gates in the presence of potassium. What that's important for, we don't yet know, but it tells us there are other functions out there we haven't studied."

What about the sensitive plant?

So are these channels at the bottom of the really fast plant movements like the sensitive plant's famous touch shyness? (To see a movie of this and other "nastic" (fast) movements, go to the Plants in Motion site maintained by Haswell's colleague Roger P. Hangartner of Indiana University).

Haswell is circumspect. "It's possible," she says. "In the case of Mimosa pudica there's probably an electrical impulse that triggers a loss of water and turgor in cells at the base of each leaflet, so these channel proteins are great candidates.
